Freshly plated human hepatocytes can be used to perform experiments where an intact cellular system is required. Intact, viable hepatocytes contain the major hepatic drug-metabolizing enzymes as well as the co-factors required to study Phase 1 and 2 reactions. Plated hepatocytes respond to prototypical enzyme inducers in a predictable manner and are useful for evaluating the enzyme-inducing potential of drug candidates.

Each order of plated hepatocytes is evaluated by experienced analysts to ensure good cell morphology and confluency. The product is accompanied by a datasheet that contains donor demographics and a photomicrograph of the culture immediately prior to shipment. Finally, we ensure quality throughout the culture period by performing in-house induction analysis, using prototypical inducers, on each individual donor. These data are provided to you within 1 week of shipment.

Fresh suspended hepatocytes are useful for short-term drug metabolism studies (less than 4 hours) and are also suitable for hepatic uptake studies. These hepatocytes are not recommended for use in evaluating the enzyme-inducing potential of drug candidates. 

Each order of fresh suspended hepatocytes is evaluated by experienced analysts to ensure a large yield of cells with a high viability. The product is accompanied by a datasheet that contains donor demographics and cell viability of the suspension immediately prior to shipment. Finally, we characterize each donor for phase I and II activity and these data are provided to you within 1 week of shipment.

Safety, Handling, and Storage

Plated Products

Plated product will be sent at ambient temperature and should be stored at 37°C at 95% humidity with 5% CO2 atmosphere.

Included with each plated order:

    •    Datasheet with demographics, donor serology and a picture of the cells prior to shipping
    •    Sterile replacement lid for each plate ordered
    •    Instructions for media replacement
    •    Recommended dosing and incubation conditions for enzyme induction assessment

Needed before you begin

    •    Personal protective equipment (gloves, lab coat, protective eyewear)
    •    Biological safety cabinet
    •    Aspirating device (i.e. pipette, vacuum aspirator)
    •    Incubator (95% humidity, 5% CO2, 37° C)
    •    Refrigerator or equivalent storage chamber (2-8° C)
    •    Prototypical inducers and DMSO (or applicable solvent control)
    •    Cell culture media with supplements (XenoTech Product Number K2300 recommended)

Suspensions

Suspension will be sent on wet ice and should be used on the day of receipt.

Included with each suspension order:

    •    Datasheet with demographics, donor serology
    •    Instructions for cell recovery and resuspension

    •    Biological safety cabinet Centrifuge (low speed)

    •    Hepatocyte Incubation Media (XenoTech Product Number K2500 recommended)

Caution

In the case of human tissue, XenoTech accepts only non-transplantable tissue from donors who test negative for HIV 1 and 2, HTLV, and Hepatitis B and C. However, as a precaution, all human-de­rived samples should be regarded as a potential bio­hazard and should be stored, handled and discarded accordingly. Cellular and subcellular fractions are intended for in vitro use only.
