bDNA Probes

The branched (bDNA) assay is a sensitive, non-PCR method for quantifying relative levels of mRNA. Our probe sets are designed to recognize the mRNAs encoding a variety of CYP and other drug-metabolizing enzymes.
The bDNA assay has the potential to provide a high-throughput assay for screening drug candidates for their enzyme-inducing effects in cultured hepatocytes. 1,2,3,4,5 A single 60-mm dish of cultured hepatocytes provides sufficient mRNA to analyze the expression of 20 or more mRNA species.
Each bDNA probe set consists of short DNA oligonucleotides designed to capture the mRNA species of interest. Grouped according to their function in the bDNA assay, namely Capture Extenders (CE), Label Extenders (LE), and Blockers (BL), these probes give a chemiluminescent signal proportional to the levels of mRNA, which are typically compared with the levels of mRNA encoded by a housekeeping gene, such as glyceraldehyde-3-phosphate dehydrogenase (GAPDH).
The oligonucleotide probes for the target mRNA are provided in 3 separate vials at concentrations recommended by the manufacturer of the bDNA kit.
Specific instructions for the use of this product are provided with the bDNA Signal Amplification Kit. Each probe set is sufficient for up to 1000 measurements of specific mRNA.
